Can the Gen Y Riser be used with a BlueOx Avail Tow Bar in order to tow a 2022 4x4 Sprinter Van behind a Class A motorhome?

Yes, you can use the Gen-Y Adjustable 2-Ball Mount w/ Stacked Receivers item # 325-GH-625 to flat tow with. This adjustable ball mount has a 21,000 lbs capacity and fits a 2-1/2" hitch. It is compatible with the Blue Ox Base Plate Kit item # BLU33MR that fit your 2022 Mercedes-Benz Sprinter 2500.
The Gen-Y adjustable mount will make a great high low adapter and the built-in 2-1/2" x 2-1/2" receiver size will work perfectly with the Blue Ox Avail Tow bar with a 2-1/2" receiver.
This set up won't work with the item # BX7420 which only has a 2" receiver. If you have the 2" receiver Avail Tow bar you will need the Gen-Y Hitch Reducer Sleeve item # 325-GH-007.
