I am interested in seeing if my 5lug utility trailer can accept a hub that supports 6lug. The purpose for this change is to allow the usage of wheels that match my truck. My struggle is how to confirm the bearing assembly have the same bearing sizing and alignment on the spindle. Any info would be greatly appreciated.

Yes, it is possible.
First you will need to find the correct hub and drum that will work with what you have currently on your trailer. You will have to match up the inner bearings and the out bearings with what you have currently. The bearing number will be stamped on them. If you can not find the number you can use a digital caliper # PTW80157 to find the size you have.
There are a couple common sizes for a 3,500 pound axle inner bearing # L68149 and outer bearing # L44649.
If this is the size you have then you can find a hub and drum that has a bolt pattern of 6 on 5-1/2. For example The Trailer Hub and Drum Assembly # AKHD-655-35-K works well with electric and hydraulic brake assemblies and comes with bearings, races, grease seal, grease cap, and lug nuts.
If there bearing do not match up with what you have please reach back out to me and I will make a correct recommendation.
