The pilot diameter refers to the center hole on a tire/wheel combo like # AM3H290. This is important if the wheel is centered on the hub as opposed to the lug nuts centering the wheel. It would also matter if the pilot hole was smaller than your existing wheels. A larger hole will be fine as long as the wheel is not hub piloted. In this case, the cart has a larger pilot hole so the smaller 2.5 inch hole may not fit.
Other factors when choosing a wheel/tire combo include the bolt pattern of the wheel and the tire size. I have included links to determining both bolt pattern and tire size for reference that should make choosing the correct replacement a lot easier.
