I have 5.3x12 tires on my trailer, If I put 145/80X12 can I use the 5.3x12 as a spare?

Yes, so long as it is just a temporary spare for an emergency you can use the 5.30-12 tire when switching to the Kenda Karrier S-Trail ST145/R12 Radial Trailer Tire # AM10130.
But in the event you have to use the spare you will want to switch to the ST145/R12 size as soon as you can to ensure your trailer's suspension wears evenly.
