Can this bike rack be mounted on top of the tire cover on my 2006 Honda CRV?

The Hollywood Racks SR1 2-Bike Carrier # HRSR1 referenced in your question is confirmed to fit the spare tire on your 2006 Honda CRV. I do not recommend leaving the cover on the spare tire when the rack is installed. This can potentially affect the fit and grip of the carrier and may lead to the rack loosening over time or undesired performance.
This carrier can secure up to 2 bikes at 35 pounds each and features an offset adjustment of 4-1/2 inches to either side. I recommend adding a Cable Lock like the etrailer.com # e98891 to secure your bikes to the rack.
If you plan on transporting any children's, woman's or alternative frame bikes, you may need an adapter bar like the Kuat # UB01 to provide a horizontal attachment point and improved ground clearance.
