The Derale Series 7000 Transmission Cooler, part # D13105, that you have referenced is a tube-fin design cooler and is a top rated transmission cooler. This Derale transmission cooler will fit inline with your factory transmission cooler.
You will want to route the transmission fluid through the factory cooler first, and then through the # D13105 cooler, and finally back to your transmission. So the Derale cooler will be added after the factory transmission cooler in the radiator. See attached photo that will show how the aftermarket cooler is installed on your vehicle.
The best location for the # D13105 cooler on your 2000 Ford F-250 would be between your vehicle grille and the air conditioning condenser. This location will give you the most airflow and allow the maximum cooling ability of the heat exchanger. You will want to measure to make sure you have clearance to install the heat exchanger. The heat exchanger measures 16-5/8 inches wide x 12-5/8 inches tall x 3/4-inch deep.
If you will need a transmission cooler that has a smaller heat exchanger, but still has the higher Class IV rating I would recommend the Derale Series 8000 Transmission Cooler, part # D13504. This cooler uses the plate-fin design which is more efficient than the tube-fin design and is smaller in size (11 inches wide x 12 inches tall x 7/8-inch deep).
When you run the aftermarket transmission cooler in-line after the original transmission cooler you will need a Derale Radiator Adapter Fitting, part # D13025 for your Ford F-250. This adapter will allow you to connect the aftermarket cooler to the factory coolers output fitting to allow you to run the coolant line from it without having to cut or flare the original fitting on the factory cooler.
I am including links to a couple of FAQ pages on transmission coolers.
