Will the roadmaster charge line keep the toad battery from draining the motorhome battery when the engine is off for extended periods, like overnight?

The Roadmaster Battery Charge Line Kit # RM-156-75 and products similar to it are designed to keep the battery of the toad vehicle topped off while being flat towed. Typically supplemental braking system draw power from the battery of the tow vehicle to operate which since it's turned off does not have the alternator keeping it charged.
These kits are not designed to work in the scenario you described which is almost in reverse. If your concern is batteries getting drained when parked overnight due to the connection of this kit you'd want to unplug the tow bar wiring when parked. This kit will seek to equalize the voltage between two batteries it's connected to so as long as the connection is made a charge might pass from one battery to the other.
