There is a possibility of 2 different problems. The first is that if you have the front wheel drive model RX350 and the factory tow package, then the Curt Custom Fit Vehicle Wiring, part # 56083 is not compatible with your 2010 Lexus RX350.
The other possibility is that the plug that you found on your 2010 Lexus RX350 still has the protective cover on the plug. If this is the case, you will just need to remove the protective cover and part # 56083 will plug right in.
If you have a front wheel drive model, you will need to use Tow Ready Custom Fit Vehicle Wiring, item # 118478, which will give you a 4-Way connector on your 2010 Lexus RX350. Part # 118478 plugs in-line with the vehicles tail light connectors and requires a power line to be ran to the positive terminal on the battery via in-line fuse holder.
I have linked a picture showing the vehicle connector with the protective cover. I have also linked instructions for the Tow Ready Custom Fit Vehicle Wiring, item # 118478.
