Will this set up work with lift blocks ? I lifted my Toyota and there’s 2 inch lift blocks on the suspension. Will this still work ?

Yes, it will but you'll need to add a 2" spacer item # AL52420. The Air Lift LoadLifter 5000 Air Helper Springs item # AL49DQ are confirmed to fit your 2023 Toyota Tacoma and have a 5,000 lbs capacity. The included brackets are custom fit and will bolt into your truck without drilling.
I recommend adding the Air Lift WirelessONE Compressor System item # AL25980 to your truck. This single path onboard compressor will allow you to adjust the PSI in your air bags from your phone.
