Question:

If I have a spray in bed liner, do I need to use the isolator tape on the rails in the bed of the truck? I know I would still use them on the bottom side.5th wheel brackets and rails for 2018 Ford F250 installation

The spray in bed liner of your 2018 Ford F-250 will protect the aluminum bed from the steel rails when you install Curt Custom 5th Wheel Installation Kit for Ford part # C16428-204 so you won't need to use the galvanic isolator tape on the bottom of the rails. You are correct that you need to use the isolator tape on the underside of the bed when you install the brackets because the underside is aluminum.
