The retaining nuts like what comes with the part # RG05-100 that you referenced can be reused.

No, they do not require lock washers. The torque specs for the 9/16" diameter U-bolts item #APUBR-9 is 65-95 ft lbs. With that amount of torque on the nuts they would not back off. What kind of trailer do you have?

Since you have a 3,500 lb axle most likely the diameter of the threaded portion of the spindle is 1 inch. You will need to verify that. And since the spindle has a hole in it for a cotter pin you would need a castle nut. For that I would recommend…
