Only if you have a spare larger than the 18" one. The Blue Ox Trailer Hitch item # BLU53UR, like all the other hitches for the 2020 Dodge Durango will only accommodate the 18" spare. If you have the larger full size one you won't be able to mount it back underneath your SUV when you install a hitch.
The Blue Ox Trailer Hitch item # BLU53UR has been confirmed to fit the 2020 Dodge Durango. This hitch is custom fit and bolts to your SUV with included hardware. It is a class III hitch with a 2" receiver, that has a 6,000 lbs towing capacity and a 900 lbs maximum tongue weight.

If you are planning on towing you will also need a wiring harness item # C56336 and a ball mount kit item # C45534.
