Hello, I have Dodge ram 2021, my trailer running lights went out, I changed the fuse 20 which was in F29 and lights cake back. In the evening when I needed to turn it on, it didn’t work again.

It sounds like there is a shorted circuit somewhere on your vehicle side that is causing the fuse to blow. The short would cause excessive power draw. It can also be that a wire is pinched on running light circuit.
You can use The Digital Multimeter with 42 inch Long Test Leads - LED Display # PT89ZR which can safely troubleshoot the vehicles electrical circuits to see where the short is. You can also test electrical components for faulty wiring and voltage.
