Yes, you can change from a 4.50-12 to a 4.80-12 trailer tire like the Kenda K353 Bias Trailer Tire - 4.80-12 - Load Range C # AM10062.
I was unable to find the rim size a 4.5-12 tire uses, but a 4.80-12 tire fits 12" x 4" rims, so if that's the size of your rim, you're set. Otherwise, you'd need a new wheel/tire combo, depending on your bolt pattern. For a 4 on 4" bolt pattern the Kenda 4.80-12 Bias Trailer Tire with 12" White Wheel # AM30620 or 5 on 4-1/2" bolt pattern the Kenda # AM30660.
If you aren't sure what your bolt pattern is, I have linked an article below which explains how to measure for it.
