Our 2022 Jayco travel trailer is wired for the second A/C unit in the bedroom but presently has a roof vent with a fan. Will this unit work in this vent?

The Advent Air Replacement RV Air Conditioner for Dometic Setup w/ Start Capacitor # ACDOM135 you mention is designed to be a replacement air conditioner for an existing air conditioner so since you're adding a new air conditioner, this will not work.
Instead, what you're going to want to check out is the:
-Advent Air RV Air Conditioner w/ Air Distribution Box and Wall Thermostat # ADV64FR.
This is the same exact main unit as the Advent Air Replacement RV Air Conditioner for Dometic Setup w/ Start Capacitor # ACDOM135 mentioned earlier, however this kit also includes the air distribution box and thermostat.
