You don't necessarily need to remove your wheel to measure your springs but you definitely need to take the weight off of them. If you're able to jack up the frame of your trailer high enough that the tires are suspended (even if it's just 1/2") that will be good enough to get a measurement for a replacement leaf spring. Leaf springs will expand a little over time and will appear longer when under a load which is why you'll need to do this.
If you let me know what your center-eye to center-eye measurement is as well as the weight capacity of your trailer axle I'll be able to help you out with a replacement from our linked selection although you'll likely need a 26" leaf spring like the part # e22YR for a 4,000 lb axle.
