The Pop & Lock Custom Tailgate Handle and Lock for the 2012 Chevrolet Colorado, part # PAL1700C, cannot accept a replacement lock cylinder of the type and size used for typical automotive keys. I spoke to my contact at Pop & Lock to confirm that this part is not compatible with standard auto lock components.
You might instead want to consider a power lock, since this will avoid the need to keep track of the additional keys. Pop & Lock offers a power custom tailgate lock that connects into the power lock wiring in your Colorado and integrates into your OEM tailgate handle, part # PAL8160.
