Question:
I am attempting to replace a rotted axle and it is definitely a drop axle. Am I correct in assuming that the is measured from the center of the axle to the center of the spindle?

Yes, you are correct. In order to correctly take this measurement you will need to measure from the center of the spindle to the center of the main axle tube. I have included a picture for reference.
