I use a 2016 GMC 2500 to pull a 12K lb travel trailer using the Equalizer hitch referenced in the subject line. Will this anti-rattle item work with this?

Yes, you can indeed use the Blue Ox Hitch Receiver Immobilizer II # BX88225 that you referenced to secure the shank of your Equal-i-zer Weight Distribution System # EQ37140ET in place. The Immobilizer will work to eliminate any rattle/movement you are getting at the hitch connection.
This is compatible with 2-1/2 inch hitch receivers. If you happen to have a 2 inch receiver you will want the Blue Ox # BX88224 instead.
