Hello, I am unsure of where I would use these reinforcement plates on a Thule Weekender I bought second-hand. Would they install directly under the cargo box? How do they help the functioning of the cargo box? Thank you.

Correct, the Replacement Plate Box Support part # TH10378 which is a direct replacement for the Thule Weekender 627 will install directly beneath the cargo box which you can see in the attached photo. The plates are designed to prevent the mounting hardware from piercing the cargo box or creating any deformities. If you do not have any of the mounting hardware for the box then you will need to obtain the below in addition to the Replacement Plate Box Support (4).
Replacement Quick On/Off Bracket part # TH04973 (4)
Replacement Carriage Bolt part # TH10403 (4)
Replacement Tie-Down Bracket part # TH10375 (4)
Replacement Wingnut part # TH04877 (8)
