If I want to add the Malone Axis roller at a later date do holes have to be drilled in the end uprights to accommodate it?

No, there is no drilling required to install any component of the Malone Axis Truck Bed and Roof Load Extender with Load Roller - 2" Hitches # MAL32VR or just the Malone Axis Load Roller for Malone Truck Bed and Roof Load Extender - 42" Long # MAL52VR.
I went ahead and attached a video that shows the pre-drilled holes on the uprights to install the rollers at timestamp 13:30.
