Good morning, does the 1/4inch fittings require Teflon tape to be applied when connecting to the regulator?

Yes, you will need teflon tape rated for use with propane.
The MB Sturgis Propane Hose Assembly - RV Type 1 w/ Back Check x 1/4" MPT - 12" # 100473-12 does not have an inverted flare in it so it will require tape on the end leading away from the propane tank.
An example of the inverted flare can be seen on the MB Sturgis Propane Pigtail - Stainless Overbraid - Type 1 x 1/4" Male Inverted Flare - 2' # 100833-24. If you look at the fitting running away from the tank you will see the flare.
If you wanted to use it then you would need an adapter like the MB Sturgis Propane Adapter Fitting - 1/4" Female Inverted Flare x 1/4" MPT # 402258-MBS.
