Can this light be grounded through the mounting holes or does the ground wire have to be hooked up?

The LED Heavy Duty Trailer License Plate Light # LPL41CB that you referenced is designed to ground to a ground wire circuit. You can ground it through the mounting hardware, but you would need to cut the white wire shorter and then strip it back so that you could insert the core of the wire between the mounting bracket of the light and the trailer frame.
