I think I might be over tighten Red Knob. My bike was dislodge from front tire loop in transit only to be saved by the back tire strap and lock cable. I think the red knob is loosening in transit. Is this possible?

It does sound like you may have overtightened or there is something wrong with the threading on your knob on your Yakima Frontloader. Replacing the knob should solve your problem.
If you have Yakima FrontLoader bike rack made after 2016 you will need # 8880766. If your Yakima FrontLoader bike rack was made prior to 2016 you will want # 8880226 instead. If the cap is not the issue then the mechanism is having the problem. Assuming that you're installing it right.
