I currently have a 4 pin installed aftermarket in my 2014 Nissan frontier pro4x. Will this part work or do I need to purchase one with the relay?

If the relays and jumper harness used with the 4 pole like # C56226 (see linked install video) are already in place behind the passenger side kick panel, then the Tow Ready # 118267 will work just fine.
If your aftermarket 4-pole simply tee'd in at the junction between the taillight assemblies and the vehicle taillight harness, you'd want to go with the complete kit like # 118281
