Is Adding a Set Screw Needed to Prevent Hitch Ball Nut from Backing Off

Question:
Is it safe to drill a 1/8inch hole in a Hitch ball nut and 1inch threaded shank so that a split pin can be inserted to prevent nut from loosening?

If done properly it's probably safe, but realistically if you tighten the nut of the hitch ball to the recommended torque spec you wouldn't have to worry about the nut backing off. Or you could use some thread locker if you really wanted to be cautious.
