I have 2016 ram 1500 and it has a 6 inch lift with size spacers do I need.

You can install suspension enhancement on your 2016 Ram 1500 that has a 6 inch lift like the Air Lift LoadLifter 5000 Air Helper Springs # AL57365 (this is correct kit for your truck) but you do need the correct size spacer like you mentioned. The correct size spacer will actually be a 4 inch spacer like part # AL52440 since 6 inch lifts typically only add 4 inches to the height of the suspension.
I attached an install video for the air bag kit on a 2016 Ram just like yours as well.
