How do I determine if this rim and tire will work on my current trailer. It currently has a 12 inch 4-80-12 rim and tire one it. It is currently 5 on 4 1/2 bolt pattern

You have already done most of the work in regards to knowing your bolt pattern and the Loadstar ST185/80D13 Bias Trailer Tire with 13" Galvanized Wheel # AM3S334 does have a 5 on 4-1/2" bolt pattern so the wheel is going to fit.
Since you mentioned you have a 4.80-12 tire and wheel - like the Loadstar # # AM30670, for example - I can tell you that tire and wheel measures 20.7" in diameter and 4.9" wide while the trailer tire and wheel you asked about, the # AM3S334, measures 24.4" in diameter and 7.2" wide, so it's quite a bit bigger.
The general rule is you want at least 3" of clearance from the top of the tire to your fender and at least 1-1/2" to 2" of clearance between the inside of the tire and your trailer's frame rail. If you take a quick measurement to see how much clearance you currently have, and compare that to the diameter and width of the larger tire, you will be able to tell if you have the proper clearance to use the Loadstar ST185/80D13 Bias Trailer Tire with 13" Galvanized Wheel # AM3S334.
