The description says this hitch will not fit with full size tire. Will this hitch fit my 2012 highlander with a 17 tire?

Yes, you will be able to keep your 17 inch spare tire as a spare on your 2012 Toyota Highlander with this Draw-Tite Hitch # 75586. I attached a picture of this hitch installed on a similar Highlander as yours with the spare also retained.
