I have a Drawtite Class 3 hitch on my 2016 Honda CRV. It has a 675lb tongue weight rating. Can I use a motorcycle hauler on this hitch to carry a 375lb motorcycle if there is no other passengers or heavy cargo in the vehicle?

I consulted the owner's manual for your 2016 CR-V, and found that the towing capacity for your vehicle is 1500 lbs.
The tongue weight capacity would be 10% of that, or 150 lbs which means that unfortunately you wouldn't be able to carry your motorcycle on the hitch installed on your vehicle.
Though your hitch has a 675 lb tongue weight capacity, you'll be limited to the capacity of the vehicle itself. It's not unusual for the hitch to have a higher capacity as the hitch manufacturers will design the hitch with a higher capacity so if there's a failure point it won't be a hitch.
Your best bet would be to use a small trailer such as the # K2MMT5X7 to carry your bike. If you decide to go that route, you'll need to install a trailer wiring harness like # 118561 if you don't already have one, a ball mount like # 2753 and a trailer ball like # 40038 for a 1-7/8 inch or # 63845 for a 2 inch ball.
