Question:
My 2008 Lexus RX 350 in FWD. What do I need to do after securing my car with the front wheels onto the dolly ? Neutral ? Key in accessory position ? Battery disconnect ?

I looked at the owner's manual for your 2008 Lexus RX 350 and did not find any information of dolly towing. Under emergency towing on page 387 it says to release the parking brake, shift the shift lever to N, and turn the engine switch to the ACC or ON position. This is what I would follow.
You may need to lock the steering wheel if it does not lock automatically depending on what the dolly requires. This can be done by wrapping the seat belt around the steering wheel by pulling it all the way out and then retracting it so it can't come undone.
