Hi, Is it possible to get the Yakima Skyline kit with a 70” jetstream bar for a 2020 Highlander with flush rail? Thanks

You sure can. You'll need the 70 inch JetStream crossbars as part # Y00427 along with the # Y53TR SkyLine towers and the # Y66VR landing pad kit to make the transition between the fixed mounts in your flush rail and the SkyLine towers. You can also add lock cores with part # Y07204.
I've linked to a demonstration video showing a JetStream roof rack being installed on a 2019 Highlander.
It is important to note that a crossbar system should never extend past the mirrors of your vehicle. Doing so is not only illegal in some municipalities, it is also extremely dangerous. For this reason, I recommend that you measure from mirror to mirror to make sure that you have enough length.
If you need to go to the next length of crossbars, that is the 60" # Y00426.
