Will the rack hold the bike if the tire goes flat while you are driving?

A very good question. The fit of the Yakima FrontLoader bike carrier # Y02103 will be looser if the tire was to go flat but it is unlikely that the bike will come out of the carrier. If you put the clamp on up near the front forks it will be harder for the bike to accidently pop out.
Another option is to use a carrier that holds the bike frame such as # S64720. If the tires were to go flat it would not matter since this carrier holds the bike frame.
