I realise the POL is left hand thread. What type of thread direction goes into the dual stage regulator?

The male inverted flare fitting of part # 100019-15 that goes into a regulator is a standard right hand thread. You're correct the POL connection is a left hand but that's the only one that's unique in that sense.
The inverted flare end fits into dual stage regulators that have the brass adapters like part # 204120-MBS. This way you don't need to use tape to make the connection with the hose.
