If I change the valve stem to a tpms, can I mount a 14 ply, g rated, 129M, 4080 lb load rated tire as long as the trailer load does not exceed the wheel load rating.

The Lionshead Aluminum Siberian Trailer Wheel - 16" x 6" Rim - 6 on 5-1/2 - Gunmetal Gray # LH78MR can definitely accept a Load Range G tire (like the Triangle ST235/80R16 Heavy-Duty Radial Tire - Load Range G # TR98VR). That said, I'm not really sure why you would want to. This tire is beefy (like 4,080 pounds of capacity at 110 psi beefy), but this wheel only has 3,200 pounds of capacity. G rated tires are pretty expensive and you really won't be getting the benefits from going this way, and you may run into some negative. They are going to be very stiff and bouncy.
Again, this doesn't mean you can't do this, but I think you would be a lot happier with something like the Kenda Karrier ST235/80R16 KR53 Radial Trailer Tire - Load Range E # KE35TJ. This has 3,520 pounds of capacity at 85 psi and will match the wheel significantly better. At least in my opinion.
Whichever way you end up going, the Americana Metal, Bolt-In Valve Stem - 1-1/4" Long # AM20906 is definitely an upgrade you should make. Metal valve stems are going to be the way to go for TPMS. And this one has up to 200 psi of capacity.
Now if you meant something like the Tuson Towable Tire Pressure Monitoring System - 4 Sensors - Brass Interchangeable Valve Stems # 335TPMS4WIVS (or the 6 sensor # 335TPMS6WIVS), that could so work. These can handle up to 203 psi and have plenty of capacity, and you would not need the aforementioned metal valve stems for this kind of TPMS.
