can you check 7 pin for voltage at pin for trailer brake using the manual activation button while trailer is not hooked up

Yes, if you slide the manual override lever of a brake controller you can then use a circuit tester like part # PTW2992 to test for voltage on the output pin which would be in the 5:00 position.
