Can you use the 30 amp RV plug, and the 30 amp twist lock at the same time?

Yes. You can use both of the 30 amp plugs on the 4500W Portable RV Generator # 333-0005 if needed, but you will only get 3650w total output spread out over the two outlets (when powered by gasoline).
You'll want to make sure that you start the generator without anything plugged in and then attach the appliances that are going to draw the heaviest load first, working your way down to whatever requires the least starting power.
