do you need to replace the spindle nut each time to hub is removed on a 6000 lb. nev-r-lube axle

You do not need to replace the Spindle Nut # 6-183 for your Nev-R-Lube axles every time that you remove the hub. The only time you need to replace it is when it isn't functional anymore. I reached out to my contact at Dexter Axle who confirmed this.
