Yes, from my research the 2019 Toyota Highlander is equipped to handle the weight of Longneck XL Trailer and the Yakima HoldUp part # Y02443-2 using the Curt Multi-Purpose ball mount part # D210 as long as your hitch is rated for the weight of these items. The trailer weight rating in your owner's manual is 1500 lbs but you are only as strong as the weakest component of your setup which is most likely your trailer hitch in your case.
Please keep in mind the Curt Multi-Purpose Ball mount part # D210 cuts your hitches tongue weight rating in half. I am not sure which hitch you have on the vehicle and the tongue weight it is rated for. For example the Curt Hitch part # C13200 for the Highlander is rated for 900 lbs of tongue weight. So the tongue weight rating is actually 450 lbs if you are using the Multi-purpose ball mount. As long as half of the tongue weight rating of the hitch you are using can handle the weight of the Yakima HoldUp (weighs 98 lbs), the bikes and the tongue weight of the fully loaded trailer you shouldn't have any issues using the Curt multi purpose ball mount.
