Will using a product like the Derale 3/8inch Hose Barb to 3/8inch Hose Barb Fitting Item # D98028, impede the flow of transmission fluid, and cause problems? Im wondering if I can use a fitting like this to install a transmission cooler on a 2008 Toyota Highlander, by splicing into the existing lines, and avoid taking off the bumper to install it.

For your 2008 Toyota Highlander, I recommend looking at the Derale Series 8000 Plate-Fin Transmission Cooler Kit # D13503. This cooler measures 11 inches wide, 8-3/4 inches tall and 7/8 inches deep. As long as this will fit between the grille and AC condenser core, this will be a great option for prolonging the life of your transmission and improving overall performance. This mounting location will provide the most airflow for maximum cooling ability.
This system includes barb inlets that will work with most vehicles that have 5/16 inch transmission lines. If you find that the inner diameter on the OEM lines in your Highlander will not accept the 11/32 inch hose barbs on the cooler, then you may be able to use the Derale Hose Barb # D98028 referenced in your question. It will not impede the flow of transmission fluid.
