I have a nissan frontier 2013 with a 4 inch lift with a 2 inch lift by a block method will I need a spacer to make it work properly?

The Firestone Ride Right Suspension kit part # F2558 that you referenced is a fit for your 2013 Nissan Frontier if the axle is the same distance from the frame as stock. That means that if you only did a block lift (body lift) you would not need spacers.
If you have a 2 inch suspension lift as well you would need the part # F2366 which is a 2 inch spacer kit.
