Do both the Two Ready and Curt wiring harnessed require putting the fuse in the RDX under the hood?

Yes, both of the Tow Ready trailer wiring harnesses you referenced, parts # 118500 and # 118569, require you to install the supplied fuse in the vehicle's fuse box located in the engine compartment.
Please note that these two harnesses are a fit for different model year ranges of the Acura RDX. The # 118569 fits 2013 and 2014 model years while the # 118500 fits 2007 through 2012 vehicles.
