How Does Optronics MCL45AB Double Bullseye Clearance/Side Marker Light Ground

Ref Optronics Double Bullseye LED Trailer Clearance or Side Marker Light - 10 Diodes - Amber Lens Item # MCL45AB This side marker is a single wire i assume is Positive? The mounting screws to the vehicle act as the negative lead?

You assume correctly, sir.
The black wire provides power, and the lamp would ground via its mounting screws as is shown in the install video I've linked for your convenience. If the lamp isn't directly connected to metal that's in direct contact with the trailer frame, you'll need to run a jumper wire from the mounting screw to the trailer frame to complete the ground circuit.
