I cannot confirm a fit of the Replacement Lock Cylinder for BOLT Toolbox Latch - Codes to Ford Side-Cut Key # BL7025636 for your spare tire cover in the bumper of your 2016 Ford F-150. I can say that it is an excellent question and would take out the issues with other locks and the lost key issue since it codes to your ignition key. This cylinder fits standard 3/4 inch hole of toolboxes.
This product does require the Retaining Nut for BOLT Tool Box Latch Lock Cylinder # BL4306752.
