Is there anything that will secure the ground wire to the metal on a trailer to ensure the ground wire will stay in contact with the trailer? Or is it best to just drill a hole in the metal and secure the ground wire by placing a screw through the metal rim on the end of it?

The best way to secure a ground to a trailer or any metal surface is to do exactly what you described.
You will want to make sure you have a ring terminal like part # DW05702-1 attached to the ground wire first. Then drill a hole in the surface you want to attach the ground to. Make sure the surface is clean and free of any solvents, grease, or dirt and securely fasten the ring terminal with a sheet metal screw.
