Will this fit a 2016 tundra 5.5 bed and do you need to drill holes?

Erickson Truck Bed Ladder Rack # EM07705 is a confirmed fit for the 2015 Toyota Tundra that has the 5-1/2-foot bed with OR without the OEM Deck Rail System. No drilling is required to install it since it uses clamps. A video on this rack is linked for you.

This rack is compatible with bed liners and toolboxes, but not with tonneau covers.
