How do they work with a roll in lock bed cover and will they fit on a chevy silverado?

The Putco Push-Up Tie-Downs # P99901 that you referenced are a universal-fit item intended to fit a broad range of trucks. Most installations can be completed without drilling.
These chrome-plated-stainless-steel tie downs install in your truck stake pockets. Please refer to the linked photo that shows these tie downs installed on a truck with a tonneau cover. I have also included the written installation instructions.
I checked the Roll.N.Lock website to review their M-Series roll-up covers for the Chevrolet Silverado. These appear to install within the truck bed rails, leaving the stake pockets open and accessible for installation of the tie-downs. As long as your Roll.N.Lock bed cover does not block access to the stake pockets then your installation should be as simple as that shown in the linked video.
