Can I run this line from my RVs regulator to a 100lb propane tank?

While we do have a hose that will do what you are needing, the Propane Hose with Back Check # 37207-30785 would not be the correct option. This is because a 100 lb. propane tank is going to require a POL tank connection instead of a type 1 connection.
Instead you will want to use the MB Sturgis Propane Supply Hose # 100202.
Or the Propane Hose - POL Pigtail # 37207-30685 if you have an auto change-over regulator because auto change-over regulators typically have a FIF fitting.
