Does it come with an actual key and lock or the master key setup which I would need to buy keys and locks for? The picture on the listing shows an actual lock

Yes! The Rhino-Rack 2500 Roof Rack DS126B does come with lock cylinders and 2 front cover keys that lock/unlock the locks on the feet. In order to have a complete setup with the rack, you also need the Custom DK Fit Kit for Naked Roofs # DK195 for your 2017 Toyota Prius v.
I went ahead and attached a photo of the parts list of the rack and highlighted the keys for you.
