Yes you can use the Adco SFS AquaShed Truck Camper Cover - 17-3/4' Long - Gray # 290-12264 while it is still mounted on your truck bed.
I suggest not driving with it installed at all. Although these covers are very durable they are designed to be stationary and can not hold up to the winds at highway speeds. It will cause damage and create an unsafe situation for you and and anyone behind you on the road.
